I've never heard of such a thing. I think it's wise if you are planning a long car trip to stop every few hours and take a break -- walk around, stretch your legs, etc, but I have never heard that you shouldn't ride in a car for more than 2 hours when pregnant.
The other issue is this -- let's say that 100 people respond and say that they've never heard it. What information will make you comfortable enough to go ahead and do it? What happens if there is no validity to this doctors claims, and you do it, and your wife miscarries? It could have nothing to do with the car ride, and yet, you might be tempted to blame yourselves for taking the risk.
Only you know your personality and temperament. But you need to think about these sorts of things before you go.
I would take the trip. I think car rides are safer than airplanes, as far as the cabin pressure and confines. You have a lot more flexibility in a car ride, to stop and take breaks. It sounds like a lovely way to have a vacation with your wife and see the US. --
